Personality: Junko Enoshima is a girl of paradoxes, self contradictory from beginning to end. She's calculating and patient, dark and brooding, high energy and childish, mercurial and short-tempered -- nothing about Junko is straightforward in the least and that's the way she likes it, thank you very much.
Despair is the keyword you need to start cracking her code, as it's the underlying motivation of literally everything that drives Junko as a person. She lives and breathes it and dedicates her entire life to bringing Despair to herself and others and will do whatever it takes to fulfil that goal, from the mundane and petty to the cataclysmic. There's no act too taboo, no idea too wild, so long as it guarantees her some Despair in return. Kyouko Kirigiri accuses her of having a "despair fetish" and that's not too far off the mark.
To Junko, Despair is the only thing in the world you can trust and rely on. Hope and optimism are pretty words, and they're equally pretty traps -- the higher up you are, the harder you hurt when you hit the ground. But when you've already hit rock bottom, you can't go any lower, can you? She claims to have been born in despair at her own birth and it's clear that her obsession with despair started at an early age - as told in Dangan Ronpa Zero, she once spent days building an elaborate sandcastle, only to destroy it, all for the Despair of seeing her hard work come to nothing.
This is an apt metaphor for Junko's behaviour and motivations in general. Having her hard work destroyed was, to Junko, inevitable -- so the least she could do was destroy the sand castle herself and have some agency over her own Despair. This extends to almost everything that Junko does -- pain and Despair as are inevitable as the tides, so the very least she can do is control it.
This tendency towards self-sabotage makes itself evident in Junko's later life as well. She's highly intelligent and patient, as the first two games of the Dangan Ronpa series show. Junko's machinations went on for years and she was very content to play the long game when it came to setting up the Mutual Killing Games. But as thorough as she was in her construction of the games, she was ultimately defeated by her classmates taking advantage of loopholes and clues that she herself had left behind for them to find -- even though she could have easily rigged the game in her favour.
This makes her at once a wildcard and yet strangely predictable -- even if you think you've managed to work out what Junko's planning, there's always the chance she could change her mind at the last minute, out of boredmon or pettiness. But at the same time, even if it seems like Junko's already won, there's still going to be a chance of overthrowing her victory. Because Junko will always leave a stray thread in her plans that, if pulled, with leave the whole setup crashing down around her ears. There is simply no fun in a game that she is guaranteed to win.
But despite all her cunning and intelligence, she's strangely childish, too -- she's not all that composed at the best of times, but when things start to head south for her, she's not above throwing a tantrum like a child who doesn't get their way; she rants and screams at the final survivors of the first game as they slowly pull apart her mystery and actually stamps her feet and cries to herself during Dangan Ronpa Zero when the murder of the student council of Hope's Peak isn't "despair-inducing" enough for her. Like a child, she swings between emotional extremes at the blink of an eye, taking on a different manner of speaking and presenting herself so often that it's a genuine shock when she actually uses her own voice and personality. During the final trials of both games, she cycles through these "personalities" with the claim that she's been isolated so long that she's "forgotten" what her real self is like.
It would be a mistake also to think that Junko is composed entirely of her worst traits (though she would very much like people to think she is). Under all her barbs and venom, Junko is deeply empathetic and sensitive to the suffering of others. She gained the love and devotion of Super High School Level Despair and the Warriors of Hope not with threats but by understanding them, their pain, and teaching them how to empower themselves with it. She's protective and loyal to the people she considers 'hers' -- in her own way, of course.
She's also fiercely protective of children and when presented with the opportunity, proves herself to be quite nurturing in... a less-than fantastic way. Rather than harm the Warriors of Hope when she finds them raw and vulnerable, she takes them in and practically adopts them, teaching them to fight back against their abusers and empower themselves.

As a note, Junko's history at a point further back than Dangan Ronpa Zero has yet to be discussed in canon, but it's pretty safe to say that her childhood was Not Great. It's noted that if it weren't for her awful upbringing, Junko wouldn't have come out as awful as she is and, that combined with her protectiveness of the Warriors of Hope in Ultra Despair Girls, seems to imply a history of parental abuse and at the very least, neglect.
Abilities: Being as she is a Super High School Level Fashionista, Junko is incredibly attractive and graceful, with a great deal of knowledge about fashion, makeup and the like. She's able to use this to her advantage to disguise Mukuro as herself so well that it takes the others some time to realize there was any difference between the two at all.
She's also a fantastic actress, managing to hide her true self so well that she was sealed inside the school with the very students that were being protected from her. She can lie, deceive and manipulate with the very best, spreading despair and planting seeds of distrust within her classmates with nothing more than her words and some vaguely ominous images of the outside world.
She's also incredibly intelligent, perhaps to the point of genius -- it's mentioned at one point that Junko was smart enough to have been able to claim any SHSL title she put her mind to. As Ryouko Otonashi, she has the title of 'Analyst', which is also incredibly apt. Junko is extremely analytical and uses this to her advantage to read and understand people to make it easier to manipulate and take advantage of them.
